Even when a bike is brand new the factory chain adjustment markings may or may not be accurate, also as bikes get older the normal wear and tear on components like swing arm bushings, wheel bearings and the chain adjusters themselves can effect the accuracy enough that checking and compensating for any misalignment is a good idea, the only way to really know is to take accurate measurements.
A small amount of misalignment at the axle can equal a much larger amount at the countershaft sprocket, the snail cam chain adjusters on my 99 XT600E while set at the same number, resulted in 1/8” to 3/16” difference in distance between the swing arm pivot and the axle center from one side of the bike to the other, causing excessive wear on the inside face of the countershaft sprocket. Correcting this required that one snail cam adjuster be adjusted a full 2 notches away from the factory markings. Trust your own measurements !
